In March 1984, she was named Playboy's Playmate of the Month, a highlight of her career. She studied film acting with R. J. Adams at The Actors Workshop, Orange County.
From 1986 to 1993, Speir appeared in several action and exploitation B-movies directed by Andy Sidaris, often alongside fellow former Playboy Playmate Roberta Vasquez. She is known for her role in the "Bullets, Bombs, and Babes" series, which ran for nine years. In 2019, she published her autobiography, The Naked Truth: The Fall and Rise of Dona Speir.
