She earned a BFA from the University of California, Santa Cruz in 1984 and an MFA from NYU's Tisch School of the Arts Graduate Acting Program in 1987.

Manheim's brother, Karl, is a law professor at Loyola Law School. Her son, Milo, was born on March 6, 2001, conceived via sperm donation from her close friend Jeffrey Brezovar.
